PNC-27

PNC-27 is a synthetic 32-residue chimeric peptide joining residues 12-26 of human p53, the HDM-2-binding segment, to a C-terminal penetratin-derived membrane-residency sequence. Investigators at SUNY Downstate, including Michl and Pincus, examined HDM-2 localisation in the plasma membranes of transformed and untransformed cell lines and colocalisation of the peptide with membrane-bound HDM-2 (Sarafraz-Yazdi et al., Proc Natl Acad Sci U S A 2010). Subsequent structural and imaging work characterised the peptide's HDM-2-bound conformation and examined transmembrane pore formation and membranolytic endpoints in cultured pancreatic carcinoma and melanoma lines alongside untransformed fibroblast controls (Sarafraz-Yazdi et al., Biomedicines 2022). Transplant experiments in murine and human-derived acute myeloid leukaemia models examined membrane HDM-2 expression, leukaemic blast and stem-cell endpoints, and normal haematopoietic stem-cell activity (Wang et al., Leukemia 2020). No human trials have been published and the material is not an approved pharmaceutical.
