PT-141 (Bremelanotide)

Bremelanotide, also designated PT-141, is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone, Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, characterised as an agonist at melanocortin receptors including MC3R and MC4R (Molinoff et al., Annals of the New York Academy of Sciences 2003). Preclinical work examined solicitational and other sexual behaviours in female rats following central melanocortin receptor stimulation (Pfaus et al., Proceedings of the National Academy of Sciences 2004). In humans, a placebo-controlled pilot recorded physiological and subjective arousal measures in premenopausal women with sexual arousal disorder (Diamond et al., Journal of Sexual Medicine 2006), and two randomised Phase 3 trials, RECONNECT 301 and 302, enrolled premenopausal women with hypoactive sexual desire disorder (Kingsberg et al., Obstetrics & Gynecology 2019). Bremelanotide acetate is licensed in the United States as a prescription product; the research-grade peptide is not that pharmaceutical product.
