Melanotan II
Melanotan II (MT-II) is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone, Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, designed at the University of Arizona (Hruby and Hadley groups) as a lactam-bridged alpha-MSH(4-10) analogue and characterised for melanotropic potency in frog- and lizard-skin bioassays (Al-Obeidi et al., Journal of Medicinal Chemistry 1989). As a non-selective melanocortin receptor agonist it has served as a rodent pharmacological tool, with food-intake endpoints examined in mouse hyperphagia models (Fan et al., Nature 1997). Human data are limited to small early studies: a pilot phase I study in three healthy volunteers recorded tolerability and skin-reflectance pigmentation endpoints (Dorr et al., Life Sciences 1996), and its development history is reviewed by its originators (Hadley & Dorr, Peptides 2006). The free-acid analogue bremelanotide is a separately licensed pharmaceutical; melanotan II itself has not been approved by any regulator.
