ARA-290 (Cibinetide)

ARA-290, assigned the international nonproprietary name cibinetide, is a synthetic linear 11-residue peptide (pGlu-Glu-Gln-Leu-Glu-Arg-Ala-Leu-Asn-Ser-Ser) whose sequence is taken from the aqueous-facing surface of helix B of erythropoietin, with an N-terminal pyroglutamate. Brines and colleagues designed it as a non-erythropoietic peptide proposed to engage a heterocomplex of the erythropoietin receptor and the beta-common receptor, and first characterised the 11-residue peptide in rodent ischaemic stroke, renal ischaemia-reperfusion and wound-healing models alongside erythropoietic-activity controls (Brines et al., Proceedings of the National Academy of Sciences 2008). A double-blind, placebo-controlled trial examined small-fibre neuropathy symptom scores and quantitative sensory thresholds in patients with sarcoidosis (Dahan et al., Molecular Medicine 2013), and a later Phase 2b trial examined corneal nerve fibre area by in vivo confocal microscopy (Culver et al., Investigative Ophthalmology & Visual Science 2017). No regulator has approved the compound and the research material is not a pharmaceutical product.
