IGF-1 LR3
IGF-1 LR3 (Long R3 IGF-I) is an 83-residue recombinant analogue of human insulin-like growth factor-I with arginine replacing glutamate 3 and a 13-residue N-terminal extension from methionyl porcine growth hormone. It was first described in Adelaide as a bacterially expressed fusion-protein analogue, with reduced IGF-binding-protein affinity and protein- and DNA-synthesis endpoints characterised in rat L6 myoblasts and H35 hepatoma cells (Francis et al., Journal of Molecular Endocrinology 1992). Nitrogen balance and muscle protein-turnover endpoints were examined alongside native IGF-I in dexamethasone-treated rats (Tomas et al., Biochemical Journal 1992). Published use centres on serum-free cell culture, where viability and recombinant-protein yield were compared against insulin in Chinese hamster ovary (Morris & Schmid, Biotechnology Progress 2000) and HEK293 cultures (Voorhamme & Yandell, Molecular Biotechnology 2006). Published human data on the analogue are essentially absent, and it is not a licensed pharmaceutical; native-sequence recombinant IGF-I (mecasermin) is a separate licensed product.
