Ipamorelin

Ipamorelin is a synthetic pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH2) developed at Novo Nordisk as an agonist at the growth hormone secretagogue (ghrelin) receptor. Initial characterisation in rat pituitary cells, anaesthetised rats and swine compared growth hormone release with GHRP-6 and GHRP-2 and measured ACTH, cortisol and prolactin as selectivity endpoints (Raun et al., European Journal of Endocrinology 1998). Rodent work examined a postoperative ileus model, recording colonic transit, faecal output and food intake after intestinal manipulation (Venkova et al., Journal of Pharmacology and Experimental Therapeutics 2009). Published clinical data comprise a pharmacokinetic-pharmacodynamic characterisation in healthy male volunteers (Gobburu et al., Pharmaceutical Research 1999) and a Phase 2 double-blind, placebo-controlled trial in bowel-resection patients (117 enrolled, 114 analysed), whose primary endpoint, time to tolerance of a solid meal, did not differ significantly between groups (Beck et al., International Journal of Colorectal Disease 2014). The compound is not an approved pharmaceutical in any jurisdiction.
