Tirzepatide
Tirzepatide (LY3298176) is a synthetic 39-amino-acid peptide developed by Eli Lilly that acts as a dual agonist of the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors; its structure is based on the native GIP sequence and carries a C20 fatty di-acid modification (Coskun et al., Molecular Metabolism 2018). The United States FDA approved tirzepatide for type 2 diabetes in 2022 (Mounjaro) and for chronic weight management in 2023 (Zepbound). Published phase 3 literature includes the SURPASS programme, which examined glycaemic parameters in participants with type 2 diabetes, and the SURMOUNT programme, which examined body-weight regulation (Jastreboff et al., New England Journal of Medicine 2022).
