Retatrutide
Retatrutide (LY3437943) is a synthetic peptide developed by Eli Lilly as a single-molecule agonist of the glucagon-like peptide-1 (GLP-1), glucose-dependent insulinotropic polypeptide (GIP), and glucagon receptors. Its discovery and receptor pharmacology have been characterised in vitro and in rodent models (Coskun et al., Cell Metabolism 2022). A phase 2 study in adults with obesity examined body-weight change and metabolic parameters over 48 weeks (Jastreboff et al., New England Journal of Medicine 2023), and the compound has since entered the phase 3 TRIUMPH registrational programme, which is evaluating it in obesity-related conditions.
