MOTS-c
MOTS-c is a 16-amino-acid mitochondrial-derived peptide encoded by a short open reading frame within the mitochondrial 12S rRNA gene, first characterised by Lee et al., Cell Metabolism 2015. Published literature has examined its interaction with AMPK signalling and folate-methionine one-carbon metabolism, largely in cultured cells and rodent models of diet-induced obesity, insulin resistance and advanced age. Exercise physiology work has measured endogenous MOTS-c expression in trained rodent tissue and after acute exertion (Hyatt et al., Physiological Reports 2022). Human evidence is limited: the MOTS-c analogue CB4211 completed a Phase 1a/1b safety and tolerability study before the programme was discontinued.
